Will this adapter fit on iMac 27 null

It fits everywhere a standard 3.5 inch hard drive fits. So if you have a regular size drive, than it will fit in it's place.

It depends on the year I guess since so much has changed over the years, but, yes, I put it in a 24 inch IMac with a HDD.

Yes, but unless you’re an experienced tech, any later model 27 inch iMac is difficult to o open and get at the hard drive bay. Which year is your iMac?
