“Do the M.2 2nd 3rd and 4th slot have a cooling pad for the rear as well as the metal cooling plate on the top? I am planning on using slot 1 for a Samsung 990 Pro 2TB and slot 2 or 3 for a WD Black SN850 4TB card.”

No there's nothing touching the back of the M.2 in the other 3 slots. Only the top hits the heat spreader.

One of the M.2 does appear to have a better heatsink than the other three. This would probably be sufficient cooling for your average M.2 SSD.
