The HDMI port on the motherboard which is not accessible is for the integrated graphics from the Ryzen processor. You always want to run your monitors off the GPU ports, which include an HDMI port. The motherboard port is blocked off simply to prevent any confusion as to which ports are for the GPU.

The RX 580 card has one HDMI port, not blocked, and three DisplayPorts. I am currently driving three monitors off the DisplayPort connectors. You can also run a multi-monitor configuration using the HDMI port with the DisplayPorts. I do not know if you can run all four connections at once, though. Hope this helps!
