(Cannot see the referenced photo) No, the adapter will allow two 2.5" SSDs to be mounted together in the same drive bay; but there is not enough room to fit a standard 1" high 3.5" HDD in there as well.  The bay is not big enough for the adapter and the 3.5" drive.
If you are asking about using two bays in the desktop, then yes, that is possible.  The adapter with the 2.5" SSD in one bay and the 3.5" HDD in the other bay.

This bracket only holds on 2.5 drive. I believe what you are asking is if you can mount another one underneath using the same bracket which is not recommended due to these going in the 3.5 slots.
