If you're referring to the banner ads on the home screen then yes. It's not an easy process, but XDA and other sites have guides to do it. Since this is Android you can install another "Launcher" that doesn't include ads instead.

Your best bet is to lookup a customer launcher app for the Shield and go that route. You cannot completely diminish or get rid of all of the ads on the home screen. You CAN create your own new home screen that is ad-less and make it the default launcher though.
