“Is my 5yr old ASUS Essentio CM6850 big enough to fit this power supply? Current PSU states "380W peak" and this is simply not enough power anymore. The power fan seems to stay on nearly all the time after I added more memory and replaced the sound card.”

Yes itll work. Also when adding upgraded stuff be sure and upgrade your PSU as the load will diminish causing you the problems. Also note on any PSU you dont always get the rated output. In effect if it says 380W peak thats when all conditions are right and normal but Voltage isnt constant. So the 380 could go down but its not a constant 380 W.

This is standard size PC/ACT form. Should fit. More power is definitely better.

I looked up your model and it has the ATX form factor required for this but be aware that the fan is inside the system blowing down. You may not have optimized cooling though the set up will work.
