The device will support your intended connection (printer's ethernet cable) since it has an ethernet port on the bottom. The issue is signal strength and the communication between the extender and the router. You indicate that the main router is in the basement and the office is on the second floor; that says that there are two building levels between them with floors and walls to deal with. Netgear specs on the range are subjective, you know (depends on bla, bla etc). The good news is that the device has a signal measuring function and it tells you via light colors your signal strength (you want green) . My take is the following: get it and plug it in the office where you can connect your printer. set as an extender and see what the light tells you; if it is green, you are home free. Connect your printer and wah..la. By the way, mine is 40 feet and three walls away and does have good signal (my house is single level).
