“I over-purchased these exercise mats. I am wondering whether I can put them all over the garage floor and still use one space, which means that I would be driving my car onto the mats, and leaving it on mats. Can they support the car being on them daily?”

I use them under almost everything and I do see that it can't take high weight. My daughter stool for her vanity sits on one and it definitely leaves indentations. It's okay for us but you will have tire prints over time.
