When you're on the primary clock screen, simply swipe down to get access to some quick access settings, including the waterproof setting (looks like 2 drops of water). The feature also enables automatically if you start the S Health swimming exercise program.

It’s always waterproof. If you mean to enable dispersing water out of the speaker, all you have to do is swipe up from the bottom and tap the little water droplet and that’s it.
