“Will the JBL GX628 Series 6.5" work with a stock stereo (no amp) of a 2000 Chevy Silverado 1500? If so, should I discard the existing separate OEM tweeter or keep the tweeter to play with the JBL speakers?”

Depends on what you're looking for. These mids will work the factory stereo just fine and the tweeters you have will give you more clarity of sound for vocals. This isn't a perfect analogy but i think of it this way, subs make you feel the music, mids let you hear the band, tweets let you hear the lyrics.
