You surely can! I am using Show, Echo & Dot in the same room, but I plan to move Echo & Dot to another room, because Show takes care of the entire room nicely.

Sorry, there was a problem. Please try again later.Actually you don't want them in the same room since both would respond to enquiry. We moved our Echo to a different room than the Show and have several Dots in different rooms. I can use the wake word and hear multiple units respond with attention tones but only the unit I'm closest to answers my request. They are autonomous units and you don't need one to use the other. However, with units in different rooms setup with the Drop In feature, if you limit it to household only, you can talk to someone in another room as you would on an intercom. It's actually pretty cool to use.
