I think remote is always connected even when fire stick in sleep mode. This remote uses the very low power Bluetooth connection so battery last long. Usually Duracell battery last 6 months. You need not to turn off remote unless you really need to stop using the fire stick for very long time. To off the remote you have to remove battery from remote.

If you don’t want to use Alexa just don’t hit the voice command button
