If you are using an Xbox One, once you have logged into your xbox unplug the USB cable and plug it back in. I don't know what the issue is with the Xbox One USB ports but once you do this it should assign your headset. Works every time for me. The other option is to flip the switch on the back of the MixAmp from Xbox to PC and back again. My understating is this is an issue with the Xbox if you don't power it completely off and use low power mode when turning it off for faster startup. The USB ports suspend and don't go completely off. This has something to do with the Xbox not seeing the mic on startup.

Did you turn switch back from PC to Xbox after you did update? Make sure the little black box on headphones is closer to your head and not the amp. If lights are red on amp. Could have the following issues. It’s on mute, headphones are no completely plugged in all the way.
