You can fly both drones simultaneously and shouldn't have too much of an issue. Radio interference is possible, but you shouldn't notice an issue.

But if you are asking, will controller A generate noise to controller B's frequency? The answer is Yes. I have been an RC flyer for almost 45 years. In the FM era, that was the case. (first question), but now they pair with each other to filter out communication exclusively to it. The second one is more Bandwidth utilization, that the higher the noise (more devices at the location), the less distance you have and probably the reaction as it has to repeat. Please note this is a very simplified answer with some basic examples.
