I doubt it. The stick's remote works off wifi (I think)

No, the stick requires RF type remote as it's mounted behind TV and out of sight. Harmony 650 is only capable of IR signal.

The Roku stick uses Direct Wi-Fi for its remote control connection vice infrared (which is what the Harmony 650 uses like most universal remotes). As such, you won't have to physically point the remote at the TV when using it.

No, the older harmony remotes cannot control the roku streaming sticks. The newer 'hub' based wifi harmony remotes work with the roku stick.
