• The smaller card had to be placed in the adapter. Once this was done, everything worked. Thanks.

• The SanDisk Ultra PLUS 32GB SDHC is compatible with the Nikon Coolpix B500. Check to make sure the card is not locked.

• I don't know about the B500, but it works with my Coolpix W100. I did have to try snapping it in place a second time-- the amera apparently didn't recognize my first effort, and used up internal camera memory instead. Also: was it a new card? If you'd used it in another device first, you probably need to "format" the card. (check instructions.)
