The best way to locate the studs would be to remove the top row of tile, locate and mark the studs then fill in the area where tile was removed with a piece of wood or plywood cut to fit the area and exactly the same thickness as the tile. This area will be covered by the microwave once it is installed. Yes it's very important to have the mounting bracket screws going into the stud, or studs. When installing the bracket screws you will be able to confirm you are properly hitting the studs as there will be resistance when the screw goes in. If it goes in too easy you're probably off the stud slightly so adjust your drilling hole to left or right to make contact.
