Yes. I cloned my W10 OS onto a larger 870 EVO using this case. I then was able to boot my machine from the cloned drive, still inside the case, attached to USB3. (You do have to boot to BIOS and change boot order to USB to do this). After this test, I went ahead and swapped the drives and reset my BIOS to boot normally.
