Yes you can. You can either download the Linksys Smart Wifi app to setup locally over the router's wifi network. Or you can use a compatible mobile web browser to use the Linksyssmartwifi.com cloud based setup. As for Comcast service, I am not familiar with them but it should not make a difference. Note that to use the linksyssmartwifi.com process the router needs to get an IP address from your (Comcast}. I assume you are looking to replace the Comcast router with this so that should not be a problem.

The router comes pre setup so you plug it in and cable it to your modem (I too have Comcast/Xfinity) - locate the SSID and use the provided password. Using the Smart Wi-Fi App (Apple also for Android) from Linksys, you can make adjustments to SSID, Passwords, guest access, channels, channel width, security type and more

This router can be configured without a computer, however, it is not advised because there are some settings that you need to configure that cannot be done using a handheld device. We would still recommend that you configure this router using a computer so that you can make the device work to its full potential.
