I did not have to turn off the water supply. I just simply twisted the filter off and snapped the new on into place. Not a single drop of water came out when I switched them out.

Thanks for the question. It is usually not necessary to turn off the water supply when changing the filter in Samsung refrigerators, though doing so may make it easier to remove the old cartridge or install the new one since it can alleviate the pressure in the lines. - Ms. Samsung
