I have the same mower (21" self propelled 80v pro) with the same batteries included. No, it does not matter which battery is inserted into the chambers. Yes, it always starts out using the left battery first then switches after left battery is depleted. This is why, after charging I switch the battery arrangement from the previous cut therefore keeping battery usage equal.
