Yes, the case has a built-in screen protector and no, it will not work with the tempered glass screen protector.

It makes it waterproof, dust proof, etc. There is a little cover that you pull open to access the charging port. I have never used a screen protector under one of these cases, but I would imagine it would not be easy to see/use the screen with two protective layers rather than just one. LifeProof has other styles that look the same and a lot of the colors match too, and they do not have a full enclosure.
