Yes, you do need the Luigi’s Mansion 3 game to be able to play the multiplayer pack.

This should fit fine on your Switch's memory, as this is NOT the full game. This is NOT a stand-alone game. I assume you have a physical cartridge of the full game and you are just buying this DLC to add-on. If you are buying the game digitally, then you may run into problems with needing more memory if you have a few games downloaded. However, you can archive large files when not in use, and download them when you do want to use them.

No. After the game updates you will be able to use the pack. If you want to play 2 player in story mode you have to unlock Gooigi.
