“Looking to get Orbi for a 4000+sqft house. Current router from Fios is in 1st floor family room on frontside of house. Wifi is weakest on 3rd floor backside of house. Thinking of keeping router in same spot, but how to set up the satellite? Wired/wireless”

You’ll want the satellite somewhere about half way between the base unit and the farthest location.

This model does support a dedicated wifi 6 backhaul channel, so in theory the router and satellites should not need to be wired. However if it's feasible for you to connect them via Ethernet, I would recommend that because it eliminates a possible point of failure, especially in a larger house.
