Yes, the D500 will autofocus while shooting video. That said, the microphones may pick up noise from the lens while in autofocus mode, so you may be better off using manual focus.

The D500 will autofocus when you have the camera's focus mode set for Full-time-servo AF - AF-F. You would choose this focus mode by pressing the AF button on the side of the camera and rotating the camera's main command dial.
