You should pick only one. The sub is self-amplified, so the connection you put into it should be line-level (unamplified) not speaker-level (amplified). Some amps know about this and provide a line-level "sub out", but you should check to be sure. If you use the RCA inputs, that will be straightforward. If you use the speaker wires, you can connect either line-level inputs or speaker-level inputs to the "input" side. I don't know why the label it "speaker level" because what comes out is NOT amplified. The sub will "strip" the subwoofer frequencies off of that signal and pass the rest of the signal on to the speakers. If the input is amplified, then the output will be speaker-level. If it's not amplified, then the output going to the speakers will still be line-level (you could connect it to an amp or to powered speakers).
