Your ears stopping up (barotrauma/airplane ear) is caused by a pressure differential between your inner ear and the environmental air pressure. Connecting the inner ear to outside air is the eustachian tube, which is supposed to keep inner and outer air pressure stabilized. But during ascent and descent sometimes the eustachian tube can't always keep open, so relatively higher air pressure trapped in your inner ear makes your ear drum hurt. More likely when you have a cold. The only way you could stop airplane ear would be with an entire pressurized suit. Headphones have nothing to do with the eustachian tube.
