“Does the laptop need to be on for the cooling pad to work. For instance will it run as needed when laptop is in sleep mode or does it continue to run at all times?”

If it's plugged into the USB port of your laptop, it will depend on your laptop if it's running or not when in sleep mode. Most laptops do not provide power to USB ports while in sleep so the fans would not be on.
