“I have a 43 in TCK Roku TV and am interested in the 65S434 android tv. If I use them both in the same room will the remotes clash or are the independent of each other?”

Roku remotes “bond” with the TV or Roku device that they’re being used with. You can have 4 Roku TVs in the same room, and they shouldn’t cross connect unless you try to pair one remote with multiple TVs powered on.

The remote will function as an IR remote until Google assistant is connected (BT), after the remote is connected through BT, then only the power button will function as IR, the rest of the buttons will work on BT.
