The Glass is not very thick so I would think that it would work with the Defender series case, but obviously if you don't remove the built in plastic screen protector from the Otterbox, you will have 2 layers of protection on the screen. I don't know how that would affect the operation of the touch screen. I had an Otterbox Defender on an iPhone 4 a few years ago, and the plastic screen protector got a few scratches that I couldn't clean off. I took the plastic piece out of the Otterbox, applied an Invisible Shield protector to the phone, and put the Otterbox back on. It worked great. I don't know why you couldn't do the same with the Glass model. In fact, most of the Defender cases that we have owned for our iPhones have left a slight unremovable ring on the front screen of the phone where the cutout is for the speaker and camera at the top. Using the Invisible Shield under the Otterbox would actually protect against that.
