You can, but the bigger question is what program are you going to use to play the files that are on that disk? I'd suggest sideloading Kodi or VLC from the appstore.

Also, with the first generation Fire TV Box you can use the external memory to store your apps etc on but you can't with the newer 4K device. Instead you will store your apps etc on a micro SD card.
