“I want to buy a WD BLACK SN750 NVMe M.2 2280 250GB which i would like to put my os in and a SAMSUNG 970 EVO M.2 2280 1TB for media. Can i use them both or there will be some conflicts? Does it matter which one is install to the top/bottom M.2 slot?Tks”

You can populate both slots, however please note that with the bottom slot in use SATA ports 5 and 6 will be disabled. https://rog.asus.com/motherboards/rog-strix/rog-strix-z490-e-gaming-model/spec

Sorry, there was a problem. Please try again later.Also be sure to change the bios M.2 setting to PCIE, it defaults to SATA on M2_1 which appears to affect M2_2 too. After I installed my SN750 500GB in that slot, the WD app was reporting only 2 lanes. Once I tweaked that setting, it came up to 4x (lanes), which is its full capability.
