The general answer is yes, it should. I have a TV in the den connected to Spectrum cable and use this to control my Samsung TV in the living room that's not connected to cable. As with you, there's no line of sight in my case but it's probably not as far apart as to a basement. That shouldn't make a difference as long as your wifi signal is strong enough. The tricky part isn't the audio/video signal; it's the IR signal for the remote. I'm not sure if the distance or Comcast DVR will cause a problem. It's easy to set up so the best thing would be to try it out and return it on the off chance it doesn't work.
