The major difference is each is made by a different company and run different software. Amazon sells the Firestick that connects to Amazon Prime and Roku sells their products to the Roku store. Each may support connected to third party source streams play video.

Any streaming device from one of the providers of streaming content can sometimes block other providers or make it less user-friendly. Roku does have it's own content now but is not it's specialty. FireSticks are easier to hack though for things like Kodi for Xbox (although that is getting added back slowly to the Xbox).
