Yes, but your fans may run at full speed. OWC makes a special cable to remedy that.

I purchased a special SATA connector that connects to the free SATA port on the motherboard so I can keep both the CD-ROM drive and the main hard disk. The SSD sits under the CD-ROM drive, and no problem with the CD-ROM fan running full speed since the temperature sensor will remain with the CD-ROM drive as usual. It does require some careful maneuvering since you also have to unscrew and loosen the motherboard and other components to access the free port, so if you are not careful, you could damage your iMac. If you want to be safe, just buy a USB 3.0 enclosure and a USB 3.0 to thunderbolt adapter (don't use the slow USB 2.0 port if you want to feel the speed). Or hire a technician.
