• Yes, however the drive that is being cloned to requires the amount of space being used by the first drive. For example, if 300GB is being used by a 1TB drive, then 300GB is needed on the 2nd 500GB drive.

• While I do not at the moment have a second hard drive in the dock, my computer reads the single as a separate drive to read and write to. Thus, I would expect if the second drive was added to the dock, the computer would assign a separate drive letter to it and read and write accordingly. Thus, it would matter not what the size of either hard drive is.
