Yes you can, but there might be performance issues with running a game off a sub hdd because of the slower read and write speeds. It's plausible but not ideal. Id recommend going with an internal hdd and expanding and upgrading your internal computer components. It would run better and be an easier setup. Source: Best Buy Computer Associate

If you read online though there is a way to trick steam into thinking that the location of Ultra 1TB External USB 3.0 Hard Drive is actually your main disk, if you search how to trick steam into running games from a different location you will find some more info.
